This article describes the development and characterization of a set of multimerized epitope tags useful for substantially enhancing the sensitivity of protein detection. These multimerized epitope tags contain up to eight-fold more copies than current available epitope tag multimers and are potentially beneficial for any assay that involving antibody detection of proteins that would benefit from enhanced sensitivity including immunostaining, biochemistry, Western blots, ELISA, flow cytometry, and expansion microscopy, among others. Proof-of-principle was demonstrated in vivo in Drosophila, but these multimerized epitope tags can be implemented in any genetically manipulable species.
