Supplementary Figure S1. mst1-L344S mutants are sensitive to Bleomycin analog Phleomycin. Cells were grown in YES media overnight at 25ºC then 5X serial dilutions were spotted onto YES plate or YES plates containing Phleomycin. Plates were incubated at 25ºC and photographed after 4 days.

Supplementary Figure S2. Examples of cells with RPA (Rad11), Rad52, and Rad54 foci after treatment with phleomycin at 25ºC. A. Rad11-CFP and Rad52-YFP foci in pku70Δ and pku70Δ mst1-L344S. B. Rad11-CFP and Rad52-YFP foci in pht1-4KR and pht1Δ. C. Rad54-GFP foci in pku70Δ and pku70Δ mst1-L344S.

Supplementary Figure S3. pku70∆ does not affect cell length at septation in mst1-L344S at 25ºC. Length at septation at 25ºC of WT, mst1-L344S, pku70∆, and pku70∆ mst1-L334S cells. The bold line in each box represents median and the filled circles represent outliers.

Supplementary Figure S4. mst1-L344S interacts genetically with genes in the checkpoint pathways. Cells were grown in YES media overnight at 32ºC then 5X serial dilutions were spotted onto YES plate or YES plates containing the indicated drugs. Plates were incubated at 32ºC and photographed after 4 days.

Supplementary Table S1. Median, first and third quantiles of cells with Rad11, Rad52 or Rad54 percentage shown in Figure 2A and 2B.

Supplementary Table S2. Strains used in this study.
