Figure S1: The RCE flanking sequences are sufficient to establish heritable gene silencing. The extent of silencing was measured by plating RCE(∆RCcons)-ade6⁺ and RCcons-ade6⁺ yeast on adenine-limiting media and counting the number of colonies with visible signs of silencing, including red, pink, and sectored phenotypes. Representative images, along with the percentage of colonies that exhibited silencing, are shown for three biological replicates of each genotype. For RCE(∆RCcons)-ade6⁺ strains, colonies that exhibited silencing were cultured overnight and plated on adenine limiting media to measure the stability of silencing.
