Table S1 provides an overview of the DamID sequencing reads and mapping rates. Table S2 contains details about the distribution of GATC sites in the female Drosophila genome. Table S3 summarizes the DNA oligomers used for cloning, DamID-seq, and PCR screening of transgenic flies. Table S4 lists all methylated GATC sites and their sequencing reads in Dam-only, Dam-Piwi(WT), and Dam-Piwi(YK) transgenic flies. The genomic coordinates were converted to the Drosophila melanogaster genome version dm6 from dm3 by UCSC LiftOver. Figure S1 shows the annotation of uniquely mapped reads in euchromatin and heterochromatin. Figure S2 shows the annotation of multi-mapped reads in euchromatin and heterochromatin. Figure S3 illustrates the number of modified GATC sites in repeat and non-repeat sequences. Figure S4 depicts methylation peaks normalized to the GATC density in genes and repeats. Reagents Table contains an overview of fly strains, plasmids, reagents, and software packages used in this work.
